[摘要]通过对不同产地藏药沙棘膏的HPLC指纹图谱分析,为沙棘膏的品种鉴定和制备工艺的研究奠定基础。实验使用AgilentzorbaxSB-C18(4.6mm×250mm,5μm)色谱柱,以乙腈-0.4%磷酸溶液为流动相梯度洗脱,流速1.0mL·min-1,柱温30℃,检测波长360nm。建立了15个批次沙棘膏的共有图谱,提取了7个色谱峰作为指纹图谱共有峰,结合对照品对其中的3个色谱峰进行了指认,分别为槲皮素、山柰素、异鼠李素;15批药材中第14批次与对照指纹图谱比较相似度小于0.9,其他批次的相似度均大于0.9。该研究首次采用HPLC建立藏药沙棘膏的指纹图谱,该方法具有良好的精密度、稳定性和重复性,为沙棘膏质量控制和评价提供依据。

HPLC fingerprint of Tibetan medicine Shaji Gao

[Abstract]This study established an HPLC fingerprint of Tibetan medicine Shaji Gao from different habitats and lay a foundation for Shaji Gao varieties identification and preparation process. The chromatographic condition was as follow: Agilent zorbax SB-C18(4.6 mm×250 mm, 5 μm)eluted with the mobile phases of acetonitrile and 0.4% phosphoric acid water in gradient mode. The flow rate was 1.0 mL·min-1, and the detection wavelength was set at 360 nm. The fingerprints of 15 batches Shaji Gao were carried out by similarity comparation, 7 chromatographic peaks were extracted as the common peaks of fingerprint, 3 peaks were identified, which were quercetin, kaempferol and isorhamnetin. The similarity degrees of 14 batches of samples were above 0.9 and 1 batch of samples was below 0.9. This is the first established fingerprint of Shaji Gao by using HPLC. This method has good precision, stability and repeatability that it could provide basis for quality control and evaluation of Shaji Gao.

沙棘系蒙古族、藏族习用药材,藏文名为达尔布。《晶珠本草》[1]记载:达尔布除肺肿瘤,化血,治培根病。《如意宝树》中说:“达尔布治消化不良,肝病”。让均多吉说:“达尔布膏治肺病”。《图鉴》中说;“达尔布味酸刺舌。治疗肺病”。《卫生部药品标准(藏药第一册)》(1995年版)中记载:沙棘膏(藏文名为“达布坎扎”)为胡颓子科植物沙棘HippophaerhamnoidesL.成熟果实的水煎膏,同时收载于《中国药典》2000年版附录。具有“清热止咳,活血化瘀,愈溃疡”的功效,主治“气管炎,消化不良,胃溃疡及闭经”[2-3]等症。现代化学及药理研究表明,沙棘膏中主要化学成分是黄酮类化合物、维生素C等[4-6],其中槲皮素、山奈素、异鼠李素为其主要的黄酮类化合物[7-10],具有止咳平喘、利肺化痰的作用、对慢性咽炎、支气管炎、咽喉肿痛、哮喘、咳嗽痰多等呼吸道系统疾病均有很好的作用。
